I have had several people reply to our email invite to attend our event saying they will be there, but they never actually click the "confirm attendance" button within the email. Is there a way to add them into the RSVP'd guest list on the back end?

The RSVP report in your email's reporting page is essentially a specialized Click Report, so there'd be no way of actually adding people to that report drilldown.
You can, however, make a contact list from that report (select-all the contacts, Actions > Add to List > Create). That way you can also manually add contacts to the list, through the Contacts page, who decide to email instead of confirming via click.
