You can use segmentation to filter out contacts who've chronically not opened your emails. You can then further manage this segment by adding it to a list, determining which contacts may still be worth keeping (such as very recent additions based on Date Added), or email that list directly to see who opens and wants to continue receiving the emails.
After removing the contacts that are too recently uploaded, or after giving time for your re-engagement email to be opened/clicked in, you can begin deleting all the remaining non-openers and the segment list. You can also export this list if you want to keep a record of it, prior to deleting the list. If you want to actually unsubscribe these contacts, you can upload the exported file as an unsubscribe import. Make sure you have advanced email permissions turned on in your account settings, if you're wanting to do this mass-unsubscribe upload.
There's also our pre-built low engagement segmentation, which you can use to re-engage those specific contacts that rarely if ever open your emails, as well make / add to the segment list for deleting or unsubscribing.
