I see this is long in the tooth and am not sure why such a simple thing has not been implemented. If you are in the event industry you want to be able to have a "master" contact list and be able to upload a "registered" list to exclude them from the master so they do not get another email telling them to register.
The current solution on importing the registration list to delete them in the master list is just plain stupid!

This is currently available through contact segmentation, via the List Membership criteria (On List > select list AND Not On List > select list). Once a segment is created, you can add it to a list on its own, email it directly, and refresh it as desired.
