When people unsubscribe, are they taken off email lists? If I send future emails, it wont send to them, right?

If a contact has unsubscribed from you, then they are removed from all of your lists, and cannot be re-added unless they explicitly sign back up. Since they aren't on any lists, they won't be sent to.
