I'd advise sending a standalone email with the options for Yes and No setup with click segmentation. Have the Yes option segmented to add contacts to a list you've created specifically for the Yes respondents, and have the No option go to a list for No respondents.
The actual link for the buttons don't need to go anywhere special. A custom landing page where you thank them for their response could be quickly setup and utilized for this purpose.
You'd then create the automations for the individual responses, with a List Join Trigger. Set one of them to trigger from list additions to the Yes list, and the other for the No list.
After activating these automations, send your standalone email so your contacts can start clicking the options, and being segmented into the proper list. While scheduling the standalone email, make sure it's setup for Resend-to-Non-Openers, or set it up for RSNO after it's been a few days since sending.
