I'm new to Constant Contact and the system that I am working with has many lists. My question is: does a CC setup need to have at least one email address or could all the entries not be in any list but be segmented instead?

While a contact doesn't technically need to be in a list to still be an active, segmentable contact in your account, you are still required to select a list when adding a contact to your account by any method.
You can send directly to segments, once they're made.
